I have a ratshack SPL meter (digital) and have just finished up some rough prototype enclosures for my CSS FR125s. If I place them in my room and take some measurments, say, every 50hz or so, would that give me an accurate graph of my response? Also, would wall treatments dramatically effect FR? Lastly, I heard that the RS meters are not accurate and need to be 'corrected'. Does someone know what those corrections are? Thanks.

The correction files are specific to the type and age of the RS meter.
Swept sine or similar measurements are far more accurate than individual sine wave tests.
Yes wall treatments can dramatically effect the in-room response.

No. First, you need more resolution in the bass and would be best choosing frequencies on a log scale.
Second, that's a real pain and open to great variability. Get a copy of Room EQ Wizard and take some up-close measurements.
